Fledgling forensic scientist here on the DNA analysis. Because they were comparing the DNA to female descendants, they were likely looking at mitochondrial DNA. Because mitochondria are only passed down through the mother they can trace lineage. But this DNA doesn't have any input from the sperm, so there is no way to distinguish the gender from this sample.
